The present invention relates to an in vitro aging model having a multilayer structure and, more specifically, to an in vitro aging model for use in tissue aging research, including an extracellular matrix with a multilayer structure and to a manufacturing method thereof. The majority of the cellular structures of the individual are organized in three dimensions, which results in complex intercellular interactions that cannot be mimicked in two-dimensional monolayer cell cultures. Therefore, in order to develop highly bio-responsive technology in various fields such as research on diseases, customized medicine, and new drug development, there is an increasing need for an in vitro model that similarly mimics the shape of a three-dimensional cell structure. Since the in vitro aging model of the present invention implements the multi-layer ECM structure as it is, the in vitro aging model can mimic the three-dimensional structure of various tissues according to the type of cells to be applied. In particular, since being similar to the ECM structure of skin tissues, the in vitro aging model is expected to be largely used in skin aging research.
