Abstract
The present disclosure relates to compositions of encapsulated cells designed to deliver proteins of interest under the control of an inducible kill switch.
Claims70
A composition comprising a population of encapsulated cells comprising a heterologous nucleic acid molecule encoding a protein of interest and an inducible kill switch.
A composition comprising a population of encapsulated cells comprising a heterologous nucleic acid molecule encoding an IL-2 and an inducible kill switch.
A composition comprising a population of encapsulated cells comprising a plurality of heterologous nucleic acid molecules encoding an IL-12 and an inducible kill switch.
A composition comprising two or more populations of encapsulated cells:
- wherein a first population of encapsulated cells comprises an inducible kill switch and a first heterologous nucleic acid molecule encoding a first cytokine;
- and
- a second population of encapsulated cells comprises an inducible kill switch and a second heterologous nucleic acid molecule encoding a second cytokine, or an antibody that binds to a checkpoint inhibitor.
A composition comprising two or more populations of encapsulated cells:
- wherein a first population of encapsulated cells comprises an inducible kill switch and a first plurality of heterologous nucleic acid molecules encoding an IL-12;
- and
- a second population of encapsulated cells comprises an inducible kill switch and a second heterologous nucleic acid molecule encoding a second cytokine, or an antibody that binds to a checkpoint inhibitor.
The composition of any of the preceding claims, wherein the cells are encapsulated with a polymeric hydrogel selected from chitosan, cellulose, hyaluronic acid, alginate, or any combination thereof.
A pharmaceutical composition comprising a composition of any one of the preceding claims.
A method of treating a subject with a disease, comprising administering to the subject the composition of any of the preceding claims.
