Abstract
The present invention relates to an antibody specifically binding to a TSHR or an antigen-binding fragment thereof, a pharmaceutical composition comprising the antibody or the antigen-binding fragment thereof, a nucleic acid molecule encoding the antibody or the antigen-binding fragment thereof, a host cell comprising the nucleic acid molecule, and related uses. In addition, the present invention relates to therapeutic and diagnostic use of the antibody or the antigen-binding fragment thereof.
Claims22
An antibody capable of specifically binding to a TSHR, or an antigen-binding fragment thereof, wherein the antibody or the antigen-binding fragment thereof comprises:
- (a) a VH CDR1 or a variant thereof, a VH CDR2 or a variant thereof, and a VH CDR3 or a variant thereof comprised in a heavy chain variable region (VH) set forth in any one of SEQ ID NOs:
- 1, 27, 29, and 30;
- and/or a VL CDR1 or a variant thereof, a VL CDR2 or a variant thereof, and a VL CDR3 or a variant thereof comprised in a light chain variable region (VL) set forth in any one of SEQ ID NOs:
- 5, 28, and 31;
- (b) a VH CDR1 or a variant thereof, a VH CDR2 or a variant thereof, and a VH CDR3 or a variant thereof comprised in a heavy chain variable region (VH) set forth in any one of SEQ ID NOs:
- 9, 32, and 34;
- and/or a VL CDR1 or a variant thereof, a VL CDR2 or a variant thereof, and a VL CDR3 or a variant thereof comprised in a light chain variable region (VL) set forth in SEQ ID NO:
- 13 or 33;
- or,
- (c) a VH CDR1 or a variant thereof, a VH CDR2 or a variant thereof, and a VH CDR3 or a variant thereof comprised in a heavy chain variable region (VH) set forth in any one of SEQ ID NOs:
- 17, 35, and 37;
- and/or a VL CDR1 or a variant thereof, a VL CDR2 or a variant thereof, and a VL CDR3 or a variant thereof comprised in a light chain variable region (VL) set forth in any one of SEQ ID NOs:
- 21, 36, and 38;
- wherein the variant has one or several amino acid substitutions, deletions or additions (e.g., 1, 2 or 3 amino acid substitutions, deletions or additions, such as conservative substitutions) compared to a sequence from which the variant is derived;
- preferably, the substitutions are conservative substitutions;
- preferably, the antibody or the antigen-binding fragment thereof comprises:
- (a) three CDRs comprised in a heavy chain variable region (VH) set forth in any one of SEQ ID NOs:
- 1, 27, 29, and 30;
- and/or three CDRs comprised in a light chain variable region (VL) set forth in any one of SEQ ID NOs:
- 5, 28, and 31;
- (b) three CDRs comprised in a heavy chain variable region (VH) set forth in any one of SEQ ID NOs:
- 9, 32, and 34;
- and/or three CDRs comprised in a light chain variable region (VL) set forth in SEQ ID NO:
- 13 or 33;
- or,
- (c) three CDRs comprised in a heavy chain variable region (VH) set forth in any one of SEQ ID NOs:
- 17, 35, and 37;
- and/or three CDRs comprised in a light chain variable region (VL) set forth in any one of SEQ ID NOs:
- 21, 36, and 38;
- and
- preferably, the three CDRs comprised in the VH and/or the three CDRs comprised in the VL are defined by the Kabat, IMGT, or Chothia numbering system.
