Abstract
Fusion constructs are described. A fusion construct contains a peptide of SEQ ID NO: 2, SEQ ID NO: 3, SEQ ID NO: 4, SEQ ID NO: 5, or SEQ ID NO: 28, fused an antibody. As compared to the peptide or protein, fusion constructs exhibits improved penetration through the BBB, and are released on the abluminal surface of the BBB, after the post-luminal surface uptake. Fusion constructs could be used in drug discovery, diagnosis, prevention and treatment of diseases.
Claims20
A fusion construct comprising a protein, or a fragment thereof, fused at a C-terminus to a peptide of SEQ ID NO:
- 2, SEQ ID NO:
- 3, SEQ ID NO:
- 4, SEQ ID NO:
- 5, or SEQ ID NO:
- 28, through a linker peptide of SEQ ID NO:
- 6.
A fusion construct comprising a protein fused at a C-terminus to a peptide of SEQ ID NO:
- 2, SEQ ID NO:
- 3, SEQ ID NO:
- 4, SEQ ID NO:
- 5, or SEQ ID NO:
- 28, directly or through a linker peptide of SEQ ID NO:
- 6, wherein the protein has a molecular weight from about 2 KDa to about 30 KDa, and degrades at a slower rate in the endothelial endosomal-lysosomal system than an identical protein that is not fused to the peptide of SEQ ID NO:
- 2, SEQ ID NO:
- 3, SEQ ID NO:
- 4, SEQ ID NO:
- 5, or SEQ ID NO:
- 28.
