The engineered IL-7 variants, and methods of use thereof are disclosed.

An engineered IL-7 polypeptide comprising:
- a helix D, a helix A, a helix B, and a helix C, wherein the helix D and helix A are linked via a DA loop sequence.
An engineered IL-7 polypeptide comprising from N-terminus to C-terminus:
- (a) a helix D sequence that is at least 80%, 85%, 90%, or 95% identical to SEQ ID NO:
- 30;
- (b) a helix A sequence that is at least 80%, 85%, 90%, or 95% identical to SEQ ID NO:
- 31;
- (c) a helix B sequence that is at least 80%, 85%, 90%, or 95% identical to SEQ ID NO:
- 32;
- and
- (d) a helix C sequence that is at least 80%, 85%, 90%, or 95% identical to SEQ ID NO:
- 33.
