Aspects of the present disclosure provide for improved compositions of bioreducible and/or hydrolysable ionizable lipid compounds, useful in the preparation of lipid nanoparticles (LNP) for the delivery of therapeutic nucleic acids and other compounds to cells. Anionic phospholipids, including a phosphatidyl-L-serine (PS) or a phosphatidylglycerol (PG) can be included in the lipid nanoparticles to increase the transfection efficiency in human dendritic cells. In some examples, the LNP compositions are vaccine compositions comprising mRNA encapsulated in lipid nanoparticles formed from an ionizable cationic lipid compound, cholesterol, an anionic phospholipid and a PEG conjugated lipid.
