Abstract
Disclosed herein include methods, compositions, and kits suitable for use in modulating immune responses and treating diseases or disorders. There are provided, in some embodiments, nucleic acid compositions (e.g., mRNA vaccines, DNA vaccines) comprising one or more polynucleotides encoding components of an antigen presentation complex (APC). The APC can comprise an APC-assembly domain, one or more antigen presentation polypeptide(s) and a presenting peptide. A plurality of APCs can be capable of self-assembling into an antigen-presenting vesicle (APV) secreted from a cell in which the APCs are expressed. There are provided, in some embodiments, populations of APVs.
Claims31
A composition, comprising:
- a nucleic acid composition comprising one or more polynucleotides encoding components of an antigen presentation complex (APC),
- wherein the components of the APC are capable of assembling into an antigen-presenting vesicle (APV) budded from a cell in which the APC components are expressed,
- wherein the APV comprises a plurality of APCs, and
- wherein the APC comprises an APC-assembly domain, one or more antigen presentation polypeptide(s) and a presenting peptide.
A composition, comprising:
- a population of an antigen-presenting vesicles (APVs),
- wherein each of the APVs comprises a plurality of antigen presentation complexes (APCs), and
- wherein the APC comprises an APC-assembly domain, one or more antigen presentation polypeptide(s) and a presenting peptide.
