Abstract
Described herein are mass spectrometers, ion manifolds, and analytical methods for high-duty cycle, tandem mass spectrometry.
Claims21
A method of detecting a biomolecule in a biological sample, said method comprising:
- (a) ionizing said biological sample to produce ions corresponding to said biomolecule;
- (b) introducing said ions into a tandem mass spectrometer;
- (c) sequentially filtering a plurality of mass isolation windows of said tandem mass spectrometer to select a subset of said ions;
- and
- (d) performing an MS/MS scan of said subset of said ions using said tandem mass spectrometer to obtain at least one MS/MS spectrum, thereby detecting said biomolecule;
- wherein said sequential filtering:
- (i) is performed at a scanning rate that provides a 1:1 mapping of said subset of said ions to said at least one MS/MS spectrum for at least 300 ions;
- and/or
- (ii) spans a mass range of at least 400 mass to charge (m/z) to 900 m/z, wherein a mass isolation window of said plurality of isolation windows is less than 5 m/z in width such that said at least one MS/MS spectrum maps to said subset of said ions corresponding to said biomolecule in said biological sample over a duration of a chromatographic peak.
