Abstract
Microbiota-associated markers of high-grade squamous intraepithelial lesions (HSIL). The present invention relates to the field of cancer diagnosis. Specifically, it relates to an improved method for the screening, diagnosis and/or monitoring of precancerous anal lesions, in particular high-degree squamous intraepithelial lesions (HSIL), in a human subject.
Claims15
An in vitro or ex vivo use of the intracellular concentration of succinyl CoA and cobalamin obtained from the microbial cells of a biological sample comprising anal microbial cells, obtained or isolated from the anal canal of a subject in need thereof, preferably a human subject, as microbiota-associated markers of high-grade squamous intraepithelial lesions (HSIL).
A method for determining a risk of a subject having precancerous anal lesions consisting of high-degree squamous intraepithelial lesions (HSIL), the method comprising:
- a. quantifying the intracellular concentration of the microbiota-associated markers succinyl CoA and/or cobalamin present in a biological sample obtained or isolated from the anal canal of the subject;
- and
- b. classifying the subject as presenting an increased or reduced risk of having precancerous anal lesions consisting of high-degree squamous intraepithelial lesions (HSIL) based on the concentration obtained in a) for each of the microbiota-associated markers or its combination.
A computer-implemented method for determining a risk of a subject having precancerous anal lesions consisting of high-degree squamous intraepithelial lesions (HSIL), comprising the following steps:
- a. receiving or storing or having access to data comprising the intracellular concentration of succinyl CoA and/or cobalamin obtained from the microbial cells of a biological sample comprising anal microbial cells;
- b. Calculating a score value based on the comparison of the intracellular concentration of succinyl CoA and/or cobalamin, or a combined score of the same, obtained from the microbial cells of a biological sample comprising anal microbial cells received or stored in accordance with step a) with one or more reference values;
- and
- c. Returning as a result a risk or an indication of whether a subject has precancerous anal lesions consisting of high-degree squamous intraepithelial lesions (HSIL) based on the calculated score value.
