Abstract
The present disclosure provides methods for re-stimulating TIL populations that lead to improved phenotype and increased metabolic health of the TILs and provides methods of assaying for TIL populations to determine suitability for more efficacious infusion after re-stimulation.

A method for expanding tumor infiltrating lymphocytes (TILs) into a therapeutic population of TILs, the method comprising:
- (a) obtaining a first population of TILs from a tumor resected from a subject;
- (b) performing a first expansion by culturing the first population of TILs in a cell culture medium comprising IL-2 to produce a second population of TILs;
- (c) performing a second expansion by contacting the second population of TILs with antigen presenting cells (APCs) and one or more of IL-2, IL-15, or IL-21, to produce a therapeutic population of TILs, wherein the second expansion is performed for at least 14 days in order to obtain the therapeutic population of TILs;
- and
- (d) harvesting the therapeutic population of TILs obtained from step (c).

A method for expanding tumor infiltrating lymphocytes (TILs) into a therapeutic population of TILs, the method comprising:
- (a) obtaining a first population of tumor infiltrating lymphocytes (TILs) from a tumor resected from the subject;
- (b) performing a first expansion by culturing the first population of TILs in a cell culture medium comprising IL-2 to produce a second population of TILs, wherein the first expansion is performed for about 11 days to about 21 days;
- (c) performing a second expansion by contacting the second population of TILs with antigen presenting cells (APCs) and one or more of IL-2, IL-15, or IL-21, to produce a therapeutic population of TILs, wherein the second expansion is performed for about 14 days to about 21 days, in order to obtain the therapeutic population of TILs;
- and
- (d) harvesting the therapeutic population of TILs obtained from step (c).
