Abstract
A fused ring compound and a preparation method therefor and a use thereof. The compound has excellent therapeutic effect on solid tumors such as gastric cancer, breast cancer, lung cancer or urothelial cancer, or hematological malignancies.
Claims14
A compound or a pharmaceutically acceptable salt, ester, stereoisomer, polymorph, solvate, N-oxide, isotopically-labeled substance, metabolite or prodrug thereof, wherein the compound has the following structure:
- wherein, R1 and R2 are each independently selected from the group consisting of hydrogen, —CN, halogen, —ORa, —NH2, —NH(C1-6 alkyl), —N(C1-6 alkyl)2, C1-6 alkyl, C2-6 alkenyl, C2-6 alkynyl, C3-6 cycloalkyl, 3- to 6-membered heterocyclyl, C6-10 aryl and 5- to 10-membered heteroaryl;
- the alkyl, alkenyl, alkynyl, cycloalkyl, heterocyclyl, aryl and heteroaryl are optionally substituted with one or more substituents selected from the group consisting of —CN, halogen, —OH, —NH2, C1-6 alkyl, C1-6 alkoxy, C2-6 alkenyl, C2-6 alkynyl, C3-6 cycloalkyl, 3- to 6-membered heterocyclyl, C6-10 aryl and 5- to 10-membered heteroaryl;
- R4 and R4′ are each independently selected from the group consisting of hydrogen, halogen, —OH, —NH2, C1-6 alkyl, halogenated C1-6 alkyl and C1-6 alkoxy, both R5 and R5′ are hydrogen;
- or, R4 and R4′, together with the carbon atom connecting R4 and R4′, form a carbon-carbon double bond or a 3- to 6-membered carbocyclic ring, and both R5 and R5′ are hydrogen;
- or, R4 and R5 are connected to form a bond, or R4 and R5, together with the carbon atom connecting R4 and R5, form a 3- to 6-membered carbocyclic ring, and R5′ and R4′ are each independently selected from the group consisting of hydrogen, halogen, —NH2, C1-6 alkyl, halogenated C1-6 alkyl and C1-6 alkoxy;
- R6 is hydrogen, C1-6 alkyl or
- R6′, R7 and R7′ are all hydrogen;
- or, R6′ and R7 are connected to form a bond, and R7′ is hydrogen;
- Ra is selected from the group consisting of H, C1-6 alkyl or C3-6 cycloalkyl;
- Ring A is selected from the group consisting of 3- to 6-membered cycloalkyl, C6-10 aryl, 5- to 10-membered heteroaryl, 3- to 6-membered heterocyclyl;
