Abstract
The present disclosure relates to protein hydrogels, to methods for their preparation and uses thereof, for example in the repair of an osteochondral defect. The protein hydrogels can comprise a crosslinked network of entangled polypeptide chains and at least one amino acid sequence capable as functioning as a biochemical cue.
Claims39
A protein hydrogel comprising a crosslinked network of entangled polypeptide chains, wherein the polypeptide chains comprise at least one amino acid sequence capable of functioning as a biochemical cue for tissue repair.
A method of preparing a protein hydrogel, the method comprising:
- denaturing a protein in an aqueous environment to produce an aqueous composition comprising overlapping polypeptide chains;
- crosslinking the polypeptide chains to produce a denatured protein hydrogel comprising a crosslinked network of entangled polypeptide chains;
- and
- optionally at least partially renaturing the denatured protein hydrogel to produce a renatured protein hydrogel comprising a crosslinked network of entangled polypeptide chains,
- wherein the protein comprises at least one amino acid sequence capable of functioning as a biochemical cue for tissue repair.
A synthetic protein comprising an amino acid sequence having at least 90% sequence identity to the amino acid sequence set forth in SEQ ID NO:10.
A synthetic protein having the amino acid sequence set forth in SEQ ID NO:10.
