Abstract
With the present disclosure there is detailed use of follistatin as a biomarker for early diagnosis and/or prediction of type 2 diabetes, liver follistatin secretion regulated by GCKR, which use is herein reported. Further, a method of composing a biomarker signature for the early prediction of type 2 diabetes in a human is herein disclosed.
Claims10
A method of determining whether a human is at risk of developing short-term, high-risk, type 2 diabetes, the method comprising:
- obtaining blood levels of follistatin and at least one additional biomarker comprising HbA1c, proinsulin, or C-peptide from a human;
- comparing the measured blood levels of follistatin and the at least one additional biomarker with a model value based on averaged blood level values from a group of humans having a high risk of developing short-term, high-risk type 2 diabetes, wherein:
- (a) short-term is within less than 10 years, and (b) the comparison involves using k-means clustering to assess type 2 diabetes progression risk levels;
- determining if the measured blood levels of follistatin and the at least one additional biomarker are higher or lower than the model value, wherein:
- (a) if the measured blood levels of follistatin and the at least one additional biomarker are higher than the model value, the human is identified as high-risk of developing type 2 diabetes;
- or (b) if the measured blood levels of follistatin and the at least one additional biomarker are lower than the model value, the human is identified as low-risk of developing type 2 diabetes;
- and
- administering to a human identified as high-risk of developing type 2 diabetes at least one medicament, wherein the medicament is Trizepatide.

A method of determining whether a human is at risk of developing short-term, high-risk, type 2 diabetes, the method comprising:
- obtaining blood levels of follistatin and at least one additional biomarker comprising HbA1c, proinsulin, or C-peptide from a human;
- comparing the measured blood levels of follistatin and the at least one additional biomarker with a model value based on averaged blood level values from a group of humans having a high risk of developing short-term, high-risk type 2 diabetes, wherein:
- (a) short-term is within less than 10 years, and (b) the comparison involves using k-means clustering to assess type 2 diabetes progression risk levels;
- selecting follistatin and at least one of HbA1c, proinsulin, or C-peptide using recursive feature elimination to construct a biomarker model for predicting type 2 diabetes risk;
- generating a diabetes risk score based on the biomarker model, wherein the diabetes risk score indicates a high, intermediate, or low risk of developing type 2 diabetes in the human;
- providing an early prediction report of the human's diabetes risk score to enable identification of high-risk individuals for potential intervention before disease onset;
- and
- administering to a human identified as high-risk at least one medicament, wherein the medicament is Trizepatide.
