Abstract
Described are methods for the fabrication of devices for delivery of a payload to a cell, including gene transfection of the cell. Such fabrication methods involve selective material deposition for control of feature properties of the device. Such device features include projections (e.g., needles) and capture site (e.g., wells) perimeters.
Claims30
A method comprising the operations of:
- a. etching a plurality of feature regions on a front side of a substrate, the substrate comprising a substrate material;
- b. depositing a feature material on the front side of the substrate;
- c. etching a portion of the deposited feature material on the front side of the substrate, thereby forming a front side surface;
- d. etching the front side of the substrate to form a plurality of wells;
- e. etching the front side of the substrate to form a plurality of ports;
- and
- f. etching a back side of the substrate to form at least one sub-array trench, and
- wherein the substrate material is different from the feature material.

A device comprising:
- a. a substrate comprising a surface;
- and
- b. a capture site, the capture site comprising:
- i. a well having a well boundary shape at the surface and having a well depth, the well configured to support a cell;
- and
- ii. a projection extending from the bottom of the well toward the surface of the substrate, and
- wherein the projection does not move relative to the substrate.
