Abstract
The present disclosure pertains to modified immune cells comprising chimeric antigen receptors (CARs) comprising one or more intracellular JAK binding domains and one or more intracellular STAT binding domains and methods of using and making immune cells comprising said CARs.
Claims107
A modified immune cell comprising a chimeric antigen receptor (CAR), wherein the CAR comprises:
- (a) an extracellular domain;
- (b) a transmembrane domain;
- and
- (c) one or more intracellular moieties comprising:
- (i) a first JAK binding domain or a portion thereof, and
- (ii) a first STAT binding domain or a portion thereof;
- wherein the modified immune cell is or comprises a stem cell, macrophage, monocyte, or dendritic cell.
A pharmaceutical composition comprising a modified immune cell of any one of the previous claims.
A chimeric antigen receptor (CAR) comprising:
- (a) an extracellular domain;
- (b) a transmembrane domain;
- and
- (c) one or more intracellular moieties comprising:
- (i) a first JAK binding domain or a portion thereof, and
- (ii) a first STAT binding domain or a portion thereof.
A nucleic acid construct comprising one or more nucleic acid sequences encoding:
- (a) an extracellular domain;
- (b) a transmembrane domain;
- and
- (c) one or more intracellular moieties comprising:
- (i) a first JAK binding domain or a portion thereof, and
- (ii) a first STAT binding domain or a portion thereof;
- wherein the nucleic acid construct encodes a chimeric antigen receptor (CAR) comprising (a) through (c).
