The present application relates to Neuroblastoma-RAS (“NRAS”) binding polypeptides. These binding polypeptide comprise a fibronectin type III (FN3) domain having one or more modified loop sequences include a modified FG loop amino acid sequence, a modified BC loop amino acid sequence, a modified CD loop amino acid sequence, a modified DE loop amino acid sequence, or a combination thereof, and optionally one or more modified beta strand sequences, wherein the one or more modified loop sequences and the one or more optional modified beta strand sequences enable selective binding to NRAS but not KRAS or HRAS. Also disclosed are conjugates that include the NRAS binding polypeptide, polynucleotides encoding the same, and methods of using these materials for treating cancer as well as screening for other agents capable of binding NRAS.
