Abstract
The present invention provides a new and improved method to prepare a stable amorphous pentacyclic triterpenoid, a composition prepared with such pentacyclic triterpenoid, and a method to mitigate effects caused by pro-inflammatory cytokines in mammals, including humans, by providing an effective dose of a pentacyclic triterpenoid that has been processed according to the present invention. The present invention also provides an effective low-dose therapeutic composition comprising a pentacyclic triterpenoid that has been processed according to the present invention.
Claims24
A process of preparing the pharmaceutical or therapeutic supplement composition of claim 9 comprising a pentacyclic triterpenoid, wherein the process comprises:
- a) blending the pentacyclic triterpenoid, and a water-soluble polymer having a glass transition temperature lower than 170° C. to obtain a mixture;
- b) melt extruding the mixture at a temperature from about 60° C. to about 180° C. to obtain an extrudate;
- c) grinding the extrudate to obtain a ground extrudate comprising a solid dispersion of an amorphous form of the pentacyclic triterpenoid;
- and
- d) formulating the solid dispersion to obtain the pharmaceutical or therapeutic supplement composition.
A pharmaceutical or therapeutic supplement composition comprising a pentacyclic triterpenoid, wherein the pharmaceutical or therapeutic supplement composition comprises a solid dispersion comprising:
- an amorphous form of the pentacyclic triterpenoid, and
- a water-soluble polymer having a glass transition temperature lower than 170° C.
