Abstract
The invention is situated in the field of cell culturing and disease modeling. More specifically, the invention relates to a method for obtaining a neuromuscular 3D organoid demonstrating myelination that is derived from stem cells. The invention further relates to a neuromuscular organoid obtained using the method of the invention which can be used to study (peripheral) neuropathic diseases, in particular Charcot-Marie-Tooth disease characterized by myelination and/or axonal defects.
Claims15
A method of obtaining a neuromuscular 3D organoid, the method comprising:
- a) Obtaining pluripotent stem cells (PSC);
- b) Culturing said PSCs in a cell culture substrate;
- c) Resuspending the cells of step b) in an organoid differentiation substrate;
- d) Seeding the resuspended cells of step c) as a non-adherent culture;
- e) Contacting the non-adherent culture of step d) with said organoid differentiation substrate until a neuromuscular 3D organoid is formed;
- f) Transferring said formed neuromuscular 3D organoid to a recipient with a low-binding surface comprising a basement membrane matrix;
- wherein during step d) to step f) said non-adherent culture is maintained on a shaker.
