An in vitro method of isolating hemogenic endothelial cells (HECs) or enriched populations of HECs, comprising isolating CD32+ cells from a population of cells derived from pluripotent stem cells.
An in vitro method of generating a population of hematopoietic cells comprising the steps of:
- i. Culturing a population of pluripotent stem cell (PSC) in a mesoderm differentiation medium and/or hematovascular differentiation medium;
- ii. Isolating CD32+ cells from the population of cells obtained from step (i) to obtain hemogenic endothelial cells (HECs) or enriched populations of HECs, and
- iii. culturing the obtained hemogenic endothelial cells (HECs) or enriched populations of HECs obtained from step (ii) in a medium that induces or promotes hematopoietic cells to produce a population of hematopoietic cells.
A substantially pure population of CD32+ HECs or a population of hematopoietic cells derived from a substantially pure population of CD32+ HECs.
