Abstract
Devices and methods incorporate mucolytic agents into a point-of-care testing device. The sample is loaded, and then the sample travels until it encounters one or more lysis agents and/or mucolytic agents. The mucolytic agent is preferably pre-loaded onto the collection device. In a preferred embodiment, the mucolytic agent is localized between the sample application zone and the conjugate zone. In embodiments with a sample compressor, one or more mucolytic agents may be pre-loaded and dried on the sample compressor, the sample collector, in various locations on the test strip, or in the running buffer.

A method for sample analysis, the method comprising:
- (a) providing a sample analysis device comprising a sample application zone and a detection zone;
- (b) transferring a blood sample to the sample application zone of the sample analysis device, wherein the blood sample comprises an extracellular target and intact cells that comprise an intracellular target;
- (c) applying a lysis agent to the blood sample under conditions sufficient to lyse the intact cells thereby releasing the intracellular target from the intact cells;
- (d) contacting the intracellular target with a first reagent to form a first detectable binding complex comprising the first reagent coupled to the intracellular target;
- (e) contacting the extracellular target with a second reagent to form a second detectable binding complex comprising the second reagent coupled to the extracellular target;
- (f) immobilizing the first detectable binding complex and the second detectable binding complex at the detection zone of the sample analysis device, wherein the detection zone comprises capture molecules that specifically bind to the intracellular target and the extracellular target;
- and
- (g) detecting the first detectable binding complex and the second detectable binding complex at the detection zone.
A method for sample analysis, the method comprising:
- (a) contacting an intracellular target obtained from a blood sample with a first reagent to form a first detectable binding complex comprising the first reagent coupled to the intracellular target, wherein the blood sample comprises a lysis agent that lyses intact cells of the blood sample to release the intracellular target from the intact cells;
- (b) contacting an extracellular target obtained from the blood sample with a second reagent to form a second detectable binding complex comprising the second reagent coupled to the extracellular target;
- and
- (c) detecting the first detectable binding complex and the second detectable binding complex.
