Abstract
The disclosure relates to an enzyme capable of mediating the integration of large DNA fragments into mammalian genomes and uses thereof. Specifically, novel enzymes that can integrate large DNA fragments into genomes are provided, including integrases as shown in SEQ ID NO: 1-7. The disclosure further relates to polynucleotides containing nucleotide sequences encoding the integrases, and host cells containing these polynucleotides. The disclosure also relates to the integrases and methods for recombining target nucleic acids into the human genome by using the integrases.
Claims15
An integrase selected from the group consisting of:
- (a) an integrase represented by any one of SEQ ID NO:
- 1-7;
- (b) a polypeptide with an amino acid sequence having at least 80%, at least 85%, at least 90%, at least 91%, at least 92%, at least 93%, at least 94%, at least 95%, at least 96%, at least 97%, at least 98%, or at least 99% sequence identity to the amino acid sequence represented by any one of SEQ ID NO:
- 1-7, and substantially retaining the biological function of any one of SEQ ID NO:
- 1-7;
- and
- (c) a polypeptide derived from (a), which is formed by substitution, deletion, or addition of one or more amino acid residues in the amino acid sequence represented by any one of SEQ ID NO:
- 1-7, and has an integrase function.
