Abstract
The present invention provides a method for producing a 3-hydroxybutyric acid/3-hydroxyhexanoic acid copolymer PHBHHx. The method implements the production of PHBHHx by the heterologous expression of PHA synthetase PhaC and an enoyl-coenzyme A hydratase PhaJ, and achieves the purpose of controlling the molar proportion of 3HHx in PHBHHx by knocking out or knocking down the coding gene of endogenous enoyl-coenzyme A hydratase FadB in a halophile and/or adjusting the composition of a related carbon source, thereby regulating the material performance of PHBHHx.
Claims20
A recombinant halophile, wherein the recombinant halophile expresses an exogenous phaC gene and/or phaJ gene, and/or, the recombinant halophile does not express a key protein of the β-oxidation cycle pathway or expresses proteins with no functions or reduced functions.
A vector, comprising:
- 1) a phaC gene and/or phaJ gene;
- and/or
- 2) an sgRNA and/or a gene encoding Cas9 protein, the sgRNA targeting an fadB gene.
