Abstract
The present disclosure provides materials and methods for cell-free expression of epitopes for immunotherapy applications. In particular, the present disclosure provides materials and methods for expressing concatenated epitopes using a cell-free protein synthesis platform for high throughput, large scale, and unbiased epitope screening and the generation of multi-epitope vaccines.
Claims15
A screening process for identifying one or more immunogenic epitopes, the process comprising:
- expressing one or more candidate immunogenic epitopes using a cell-free protein synthesis (CFPS) expression cassette, wherein the CFPS expression cassette includes a polynucleotide encoding a signal peptide including two or more concatenated epitopes, wherein the two or more concatenated epitopes are different epitopes, a heterologous promoter upstream of the signal peptide including two or more concatenated epitopes, a polynucleotide encoding an N-terminal peptide tag and a polynucleotide encoding a C-terminal peptide tag;
- binding the one or more expressed candidate immunogenic epitopes to a substrate;
- introducing one or more antibodies to the substrate containing the bound one or more expressed candidate immunogenic epitopes to a screen for a response thereto by the one or more expressed candidate immunogenic epitopes.
