The invention provides multispecific molecules that are capable of binding to an antigen-specific T cell receptor (TCR) expressed on a surface of a cell, said multispecific molecules comprising a first molecule and a second molecule, wherein the first molecule is a polypeptide comprising (i) a peptide (p) presented in the context of a major histocompatibility complex (MHC) molecule (pMHC complex), and (ii) a first multimerization domain, and the second molecule is a polypeptide comprising (i) a domain that specifically binds a molecule expressed on the surface of the cell expressing the TCR, and (ii) a second multimerization domain, and the use of such molecules for modulating T cell activity and treating diseases such as cancers, infections, and autoimmune disorders.
A multispecific molecule that is capable of binding to an antigen-specific T cell receptor (TCR) expressed on a surface of a cell, said multispecific molecule comprising a first molecule and a second molecule, wherein:
A multispecific carrier that is capable of binding to an antigen-specific T cell receptor (TCR) expressed on a surface of a cell, said multispecific carrier comprising a plurality of first molecules and a plurality of second molecules, wherein:
A single chain peptide-major histocompatibility complex (pMHC) fusion molecule that is capable of binding to an antigen-specific T cell receptor (TCR) expressed on a surface of a cell comprising (i) a peptide (p) presented in the context of an MHC molecule (pMHC complex) and (ii) a domain that specifically binds a molecule expressed on the surface of a carrier.
A single chain peptide-major histocompatibility complex (pMHC) fusion molecule carrier that is capable of binding to an antigen-specific T cell receptor (TCR) expressed on a surface of a cell, said multispecific carrier comprising a plurality of single chain major histocompatibility complex (MHC)-peptides, wherein each single chain major histocompatibility complex (MHC)-peptide comprises a peptide (p) presented in the context of a major histocompatibility complex (MHC) molecule (pMHC complex), and a domain that specifically binds a molecule expressed on the surface of the carrier.
A method of engineering a population of cells to express at least two separate transmembrane polypeptides to modulate an activity an activity or survival of a cell expressing a T cell receptor (TCR), comprising:
A nucleic acid molecule encoding the multispecific molecule encoding a first molecule comprising a peptide (p) presented in the context of a major histocompatibility complex (MHC) molecule (pMHC complex).
A nucleic acid molecule encoding the multispecific molecule encoding a second molecule comprising a domain that specifically binds a molecule expressed on the surface of the cell expressing the TCR.
