Abstract
The invention relates to nucleic acid nanostructure delivery compositions for non-viral delivery, and methods therefor. More particularly, the invention relates to nucleic acid nanostructure delivery compositions, such as DNA origami compositions, for the delivery, for example, of small molecule therapeutics, and methods therefor.
Claims14
A method of delivering and releasing talabostat mesylate into target cells, said method comprising
- administering a composition comprising a non-viral delivery vehicle to a subject having said target cells, wherein said non-viral delivery vehicle comprises a
- a single stranded DNA scaffold comprising a single stranded DNA and a plurality of single stranded oligonucleotide staples, wherein the staples fold the scaffold into a cuboid shape, and
- talabostat mesylate, wherein said talabostat mesylate is loaded onto said cuboids without causing structural changes to the cuboid, and said talabostat is released into said target cells upon contact and uptake by the target cells.
A method of delivering and releasing talabostat mesylate into target cancer cells, said method comprising
- administering a composition comprising a non-viral delivery vehicle to a subject having cancer cells, said vehicle comprising
- a single stranded DNA origami scaffold and a plurality of single stranded staple oligonucleotides, wherein the staple oligonucleotides fold the single stranded DNA origami scaffold into a cuboid shape;
- a cell targeting peptide bound to the exterior surface of the non-viral delivery vehicle;
- a plurality of PEG-poly-L-lysine polymers, said PEG-poly-L-lysine polymers linked to the exterior surface of the non-viral delivery vehicle;
- and
- talabostat mesylate, wherein said talabostat mesylate is electrostatically loaded onto the cuboid shaped non-viral delivery vehicle without causing changes to the cuboid shape, and is released in a therapeutically effective amount into said target cells upon contact and uptake by the target cancer cells.
A method of treating a patient with cancer, the method comprising administering to the patient a composition comprising a non-viral delivery vehicle, wherein said non-viral delivery vehicle comprises a
- a single stranded DNA scaffold comprising a single stranded DNA and a plurality of single stranded oligonucleotide staples, wherein the staples fold the scaffold into a cuboid shape, and
- talabostat mesylate, wherein said talabostat mesylate is loaded onto said cuboids without causing structural changes to the cuboid, and said talabostat is released into the cancer cells of the patient upon contact and uptake by the cancer cells in a therapeutically effective amount.
