Abstract
Provided herein are systems and methods for molecule delivery into cells. The method may include steps of forming a solution comprising an amount of microbubbles, an amount of soluble molecules, and one or more cells, flowing the solution through a microfluidic device, and sonicating the solution for a period of time sufficient to form pores in the one or more cells. In particular, such methods may be used for preservation of cells through delivery of a preservation agent.
Claims18
A system for molecule delivery into cells, comprising
- a microfluidic device comprising
- a first channel for housing one or more microbubbles,
- a second channel for housing soluble molecules, the second channel in fluid communication with and intersected by the first channel, and
- a third channel for housing a cell solution, the third channel in fluid communication with and intersected by the second channel;
- and
- an ultrasound transducer positioned below the third channel or an interdigital transducer positioned on opposite sides of the third channel.
A microfluidic device, comprising
- a first channel for housing one or more microbubbles;
- a second channel for housing soluble molecules, the second channel in fluid communication with and intersected by the first channel;
- and
- a third channel for housing a cell solution, the third channel in fluid communication with and intersected by the second channel.
A method for the preservation of cells, comprising:
- forming a solution comprising an amount of microbubbles, an amount of preservation agent, and one or more cells;
- and
- sonicating the solution for a period of time sufficient to form pores in the one or more cells which permit entry of the preservation agent into the one or more cells.
A method for molecule delivery into cells, comprising:
- passing a fluid comprising an amount of microbubbles, an amount of soluble molecules, and one or more cells through a microchannel one cell at a time;
- and
- sonicating the cells as the cells pass through the microchannel such that the microbubbles form pores in the one or more cells through which one or more of the soluble molecules enter the one or more cells.
