Abstract
A cell for production of a saccharide wherein synthesis of lactobionic acid in the cell is rendered less functional or is knocked out. The disclosure further provides use of the cell in a cultivation or incubation. The disclosure also describes methods for the production of a saccharide using the cell as well as the purification of the saccharide.
Claims28
-28. (canceled)
A cell capable of synthesizing and/or that synthesizes lactobionic acid (4-O-β-galactopyranosyl-D-gluconic acid, LBA), a modified form of LBA, a glycosylated form of LBA, fucosylated LBA and/or sialylated LBA in the presence of lactose in the cell's cultivation or incubation medium,
- wherein the cell's synthesis of the LBA, modified form of LBA, glycosylated form of LBA, fucosylated LBA and/or sialylated LBA is obtained by expressing at least one gene selected from the group consisting of a gene encoding carbohydrate oxidase, a gene encoding dehydrogenase, a gene encoding lactonase, a gene encoding oxygen-dependent FAD-linked oxidoreductase, and a gene encoding pyrroloquinoline quinone (PQQ) oxidoreductase,
- wherein the cell is genetically engineered to produce a saccharide,
- wherein the cell has a pathway for producing the saccharide,
- wherein the synthesis of the LBA, modified form of LBA, glycosylated form of LBA, fucosylated LBA and/or sialylated LBA in the cell is rendered less functional or is knocked out by rendering less functional or knock out of the at least one gene.
