Abstract
Genetically modified legume plants comprising a reversibly-sterile phenotype or an improved plant-pollinator phenotype are disclosed. Methods of using the plants to produce hybrid legume seed are also disclosed.
Claims85
A genetically modified legume plant (Fabaceae) cell, legume plant, part thereof, or legume plant seed thereof comprising a reversible sterility phenotype, the genetically modified plant comprising:
- a. a sterility expression construct comprising a promoter operably linked to a sterility polynucleotide encoding a sterility factor;
- b. a fertility restorer expression construct comprising a promoter operably linked to a fertility restorer polynucleotide encoding a fertility restorer;
- or
- c. (a) and (b);
- wherein expression of the sterility factor in the plant causes the plant to be sterile and wherein expression of the fertility restorer in the plant of (b) neutralizes, blocks, offsets, prevents, or inhibits the sterility factor causing the plant to be fertile.
The genetically modified plant of any of the preceding claims, wherein the sterile plant is male-sterile.
One or more expression constructs for generating a legume plant, plant part, or plant cell thereof, comprising a reversible sterility phenotype, the one or more expression constructs comprising:
- a. a sterility expression construct comprising a promoter operably linked to a sterility polynucleotide encoding a sterility factor;
- and
- b. a fertility restorer expression construct comprising a promoter operably linked to a fertility restorer polynucleotide encoding a fertility restorer.
A genetically modified legume plant (Fabaceae), part thereof, or plant cell thereof, comprising an improved plant-pollinator floral phenotype, the genetically modified plant comprising a genetic modification of at least one target nucleic acid sequence in a polynucleotide encoding a floral trait, wherein the genetic modification modifies expression of the floral trait, wherein the modified expression of the floral trait improves a plant-pollinator floral phenotype when compared to a plant that does not comprise the genetic modification, wherein the improved plant-pollinator floral phenotype increases rate of cross-pollination of the genetically modified plant with a second plant when compared to the rate of cross-pollination between wild type plants.
The genetically modified plant of any of the preceding claims, wherein the genetic modification modifies expression of a polynucleotide encoding a large petal phenotype.
The genetically modified plant of any of the preceding claims, wherein the genetic modification is in a polynucleotide encoding a sweet nectar phenotype.
The genetically modified plant of any of the preceding claims, wherein the genetic modification modifies expression of a polynucleotide encoding a purple petal phenotype.
An engineered nucleic acid modification system for introducing in a plant a genetic modification in a polynucleotide encoding a plant-pollinator floral phenotype, the system comprising a nucleic acid expression construct comprising:
- a. a programmable transcription regulation system targeted to a nucleic acid sequence in a polynucleotide encoding the plant-pollinator floral phenotype;
- b. an expression construct comprising a promoter operably linked to a polynucleotide encoding a plant-pollinator floral phenotype operably linked to a promoter;
- or
- c. (a) and (b),
- wherein the programmable transcription regulation system, the expression construct, or both modify expression of a polynucleotide encoding a plant-pollinator floral phenotype, thereby increasing attraction of a pollinator to a plant, thereby resulting in an increased rate of cross-pollination.
One or more expression constructs for introducing a genetic modification into a legume plant (Fabaceae) of at least one target nucleic acid sequence in a polynucleotide encoding a floral trait, wherein the genetic modification modifies expression of a polynucleotide encoding a plant-pollinator floral phenotype, wherein the modified expression of the floral trait improves a plant-pollinator floral phenotype when compared to a plant that does not comprise the genetic modification, wherein the improved plant-pollinator floral phenotype increases rate of cross-pollination of the genetically modified plant with a second plant when compared to the rate of cross-pollination between wild type plants.
A genetically modified legume plant (Fabaceae), part thereof, or plant cell thereof, the genetically modified plant comprising a genetic modification in a polynucleotide encoding a floral trait selected from the group consisting of a purple petal phenotype, a large petal phenotype, a sweet nectar phenotype, and any combination thereof;
- wherein the genetic modification modifies expression of a floral trait, wherein the modified expression of the floral trait improves a plant-pollinator floral phenotype when compared to a plant that does not comprise the genetic modification, wherein the improved plant-pollinator floral phenotype increases rate of cross-pollination of the genetically modified plant with a second plant when compared to the rate of cross-pollination between wild type plants.
A genetically modified legume plant (Fabaceae), part thereof, or plant cell thereof, comprising an improved plant-pollinator floral phenotype and a reversible sterility phenotype, the genetically modified plant comprising:
- a. an introduced genetic modification in a polynucleotide encoding a plant-pollinator floral phenotype selected from the group consisting of a purple petal phenotype, a large petal phenotype, a sweet nectar phenotype, and any combination thereof and a reversible sterility phenotype, wherein the plant further comprises:
- i. a sterility expression construct comprising a promoter operably linked to a sterility polynucleotide encoding a sterility factor;
- ii. a fertility restorer expression construct comprising a promoter operably linked to a fertility restorer polynucleotide encoding a fertility restorer;
- or
- iii. (i) and (ii);
- wherein the genetic modification of (a) increases rate of cross-pollination of the genetically modified plant with a second plant when compared to the rate of cross-pollination between wild type plants;
- wherein expression of the sterility factor in the plant causes the plant to be sterile and wherein expression of the fertility restorer in the plant of (i) or (iii) neutralizes, blocks, offsets, prevents, or inhibits the sterility factor causing the plant to be fertile;
- and wherein the combination of the improved plant-pollinator floral phenotype and the reversible sterility phenotype create a synergistic cross pollination phenotype.
A method of producing hybrid seed of a legume plant, the method comprising:
- d. planting seeds of genetically modified plants comprising an improved plant-pollinator floral phenotype;
- and
- e. allowing the plants to pollinate to thereby produce the hybrid seed.
A method of synergistically producing hybrid seed of a legume plant, the method comprising:
- a. planting seeds of a sterile genetically modified plant comprising reversible sterility phenotype and an improved plant-pollinator floral phenotype and seeds of a fertility restorer genetically modified plant comprising an improved plant-pollinator floral phenotype;
- b. allowing the seeds to germinate and grow into plants;
- and
- c. allowing the fertility restorer plants to pollinate the sterile plants to thereby produce the hybrid seed on the sterile plant;
- wherein a combination of the improved plant-pollinator floral phenotype and the reversible sterility phenotype create a synergistic cross pollination phenotype.
