Abstract
Disclosed herein are plants, plant seeds, and plant cells that are modified to overexpress a trans-Golgi network (TGN) associated and microtubule (MT)-binding protein, TGNap-1. Such plants, plant seeds, and plant cells can be, but are not limited to, Arabidopsis thaliana, grape, cotton, soybean, tomato, potato, rice, sorghum, corn, coffee, wheat, citrus (e.g. orange), an apple. The modified plants exhibit approximately a five-fold reduction against the plant pathogen Pseudomonas syringae DC3000, a rod-shaped, gram-negative bacterium. The modified plants also exhibit normal plant size and biomass accumulation as compared to a control plant.
Claims20
A transgenic plant, plant seed, or plant cell comprising an expression cassette comprising a promoter operably linked to a nucleic acid segment encoding a TGNap1 protein with at least 90% sequence identity to any of SEQ ID NO:
- 1-13 or an allelic variant thereof, wherein TGNap1 is overexpressed as compared to a control plant.
A transgenic plant, plant seed, or plant cell comprising a heterologous promoter operably linked to a nucleic acid segment encoding an endogenous TGNap1 protein with at least 90% sequence identity to any of SEQ ID NO:
- 1-13 or an allelic variant thereof, wherein TGNap1 is overexpressed as compared to a control plant.
A method comprising cultivating a seed or plant comprising an expression cassette comprising a promoter operably linked to a nucleic acid segment encoding a TGNap1 protein with at least 90% sequence identity to any of SEQ ID NO:
- 1-13 or an allelic variant thereof, wherein TGNap1 is overexpressed as compared to a control plant;
- and/or comprising a heterologous promoter operably linked to a nucleic acid segment encoding an endogenous TGNap1 protein with at least 90% sequence identity to any of SEQ ID NO:
- 1-13 or an allelic variant thereof, wherein TGNap1 is overexpressed as compared to a control plant.
The method of claim 17, further comprising harvesting cotton from the cotton plant or obtaining seed from the Arabidopsis thaliana, grape, cotton, soybean, tomato, potato, rice, sorghum, corn, coffee, wheat, citrus (e.g. orange), or apple plant.
