Provided are methods for metabolically enhancing chimeric antigen receptor (CAR)-T cell function and persistence, wherein the enhancing of CAR-T cells can be used for the enhancement of CAR-T cell efficacy in cancer immunotherapy, and provided are products of manufacture and kits comprising CAR-T cells genetically altered to have greater than wild type or substantially more expression or activity of decaprenyl-diphosphate synthase subunit 2 (PDSS2). Provided are methods for metabolically enhancing chimeric antigen receptor (CAR)-T cell function and persistence comprising administering a CAR-T cell with a heterologous gene expressing PDSS2. Provided herein are CAR-T cells that have been treated in vivo or ex vivo with a nucleic acid to increase the activity of a PDSS2 in the CAR-T cell. Provided are methods for metabolically enhancing CD8 T cells resulting in their having improved metabolic fitness and increased accumulation in vivo in response to infection and tumors.
