Abstract
The present invention relates to mRNA sequences usable as mRNA-based vaccines against infections with influenza viruses. Additionally, the present invention relates to a composition comprising the mRNA sequences and the use of the mRNA sequences or the composition for the preparation of a pharmaceutical composition, especially a vaccine, e.g. for use in the prophylaxis or treatment of influenza virus infections. The present invention further describes a method of treatment or prophylaxis of infections with influenza virus using the mRNA sequences.
Claims30
A pharmaceutical composition comprising:
- (i) a first mRNA comprising a first coding sequence encoding a HA3 antigen from an influenza A virus, wherein said first coding sequence is at least 85% identical to the HA3 antigen coding sequence of SEQ ID NO:
- 213625;
- (ii) a second mRNA comprising a second coding sequence encoding a HA1 antigen from an influenza A virus, wherein said second coding sequence is at least 80% identical to the HA1 antigen coding sequence of SEQ ID NO:
- 213561;
- and
- (iii) a third mRNA comprising a third coding sequence encoding a HA antigen from an influenza B virus, wherein said third coding sequence is at least 80% identical to the HA coding sequence of SEQ ID NO:
- 213680;
- wherein the first, second, and third mRNAs each comprise, from 5′ to 3′:
- (a) a 5′-CAP structure comprising m7G;
- (b) a 5′-untranslated region (UTR) element;
- (c) the coding sequence;
- (d) a 3′-UTR element;
- and (e) a poly (A) sequence comprising 60 to 250 consecutive adenosines,
- wherein the first, second, and third mRNAs are formulated with a cationic carrier.
A method for stimulating an anti-influenza immune response in a mammalian subject comprising administering to the subject an effective amount of a composition comprising:
- (i) a first mRNA comprising a first coding sequence encoding a HA3 antigen from an influenza A virus, wherein said first coding sequence is at least 85% identical to the HA3 antigen coding sequence of SEQ ID NO:
- 213625;
- (ii) a second mRNA comprising a second coding sequence encoding a HA1 antigen from an influenza A virus, wherein said second coding sequence is at least 80% identical to the HA1 antigen coding sequence of SEQ ID NO:
- 213561;
- and
- (iii) a third mRNA comprising a third coding sequence encoding a HA antigen from an influenza B virus, wherein said third coding sequence is at least 80% identical to the HA coding sequence of SEQ ID NO:
- 213680;
- wherein the first, second, and third mRNAs each comprise, from 5′ to 3′:

- (c) the coding sequence;
- (d) a 3′-UTR element;
- and (e) a poly (A) sequence comprising 60 to 250 consecutive adenosines,
- wherein the first, second, and third mRNAs are formulated with a cationic carrier,
- wherein the composition is administered by intradermal or intramuscular injection.
A method for producing a pharmaceutical composition comprising:
- (I) obtaining:
- (i) a first mRNA comprising a first coding sequence encoding a HA3 antigen from an influenza A virus, wherein said first coding sequence is at least 85% identical to the HA3 antigen coding sequence of SEQ ID NO:
