Abstract
The technology provided herein relates to multiplex methods and kits for detecting different analytes in a sample in parallel by sequential signal-encoding of said analytes, as well as in vitro methods for screening, identifying and/or testing a substance and/or drug and in vitro methods for diagnosis of a disease, and an optical multiplexing system.
Claims20
A kit for assigning coded fluorescence patterns to a plurality of target analytes in a sample, the kit comprising:
- a first decoding oligonucleotide probe population;
- a second decoding oligonucleotide probe population;
- a first translator connector element oligonucleotide probe population whose members comprise a first fluorophore;
- a second translator connector element oligonucleotide probe population whose members comprise a second fluorophore;
- wherein the first decoding oligonucleotide probe population and the second decoding oligonucleotide probe population comprise probes that share a common identifier element binding region;
- wherein the common identifier region corresponds to a single population of an analyte among the plurality of target analytes in the sample;
- wherein the common identifier region does not directly bind to any target analyte among the plurality of target analytes in the sample;
- wherein the first decoding oligonucleotide probe population comprises probes that share a common first recruiting segment that recruits, by reverse complementary base pairing, members of the first translator connector element population when the first decoding oligonucleotide probe population is contacted to the sample;
- wherein the second decoding oligonucleotide probe population comprises probes that share a common second recruiting segment that recruits, by reverse complementary base pairing, members of the second translator connector element population when the second decoding oligonucleotide probe population is contacted to the sample.
