Abstract
The present invention relates to antibody-drug conjugates, wherein the antibody specifically binds to folate receptor α, and wherein the drug is preferably chosen among a cytotoxic drug. Such antibody-drug conjugates are useful in particular in treating proliferative diseases including cancers, such as ovarian, breast and non-small cell lung cancers.

An antibody-drug conjugate of Formula (I):
- wherein:
- Ab is an anti-folate receptor alpha (FRα) antibody which binds specifically to the folate receptor alpha of SEQ ID NO:12;
- L is a cleavable linker moiety bonded to said anti-folate receptor alpha (FRα) antibody;
- D is an inhibitor of topoisomerase I bonded to L;
- and
- p is from 1 to 8.
The antibody-drug conjugate of claim 1, wherein the Ab is an anti-FRα antibody which comprises a variable heavy chain polypeptide comprising HCDR1 of SEQ ID NO:1, HCDR2 of SEQ ID NO:2, and HCDR3 of SEQ ID NO:3 and a variable light chain polypeptide comprising LCDR1 of SEQ ID NO:4, LCDR2 of SEQ ID NO:5 and LCDR3 of SEQ ID NO:6.
The antibody-drug conjugate of claim 1, wherein the Ab is an anti-FRα antibody which comprises a heavy chain of SEQ ID NO:16 and a light chain of SEQ ID NO:17.
