Formulations Of Apremilast

A method of treating a patient suffering from psoriasis, psoriatic arthritis, or Behcet's disease comprising administering once daily to the patient an oral dosage form comprising

• a core tablet comprising:
• (a) a drug layer comprising (i) an amorphous solid dispersion of apremilast and hypromellose acetate succinate (HPMCAS) and (ii) a water soluble polymer;
• and
• (b) a swellable layer comprising one or more swellable polymers;
• a coating layer disposed on the core tablet;
• and
• a drug release orifice,
• wherein the amorphous solid dispersion comprises apremilast and HPMCAS in an apremilast:
• HPMCAS weight ratio of 40:60 to 60:40, and the amount of apremilast administered is 55 mg.

A method of treating a patient suffering from psoriasis, psoriatic arthritis, or Behcet's disease comprising administering once daily to the patient two oral dosage forms, each dosage form comprising

• a core tablet comprising:
• (a) a drug layer comprising (i) an amorphous solid dispersion of apremilast and hypromellose acetate succinate (HPMCAS) and (ii) a water soluble polymer;
• and
• (b) a swellable layer comprising one or more swellable polymers;
• a coating layer disposed on the core tablet;
• and
• a drug release orifice,
• wherein the amorphous solid dispersion comprises apremilast and HPMCAS in an apremilast:
• HPMCAS weight ratio of 40:60 to 60:40, and the apremilast is present in each oral dosage form at an amount of 27.5 mg.

An oral dosage form comprising:

• a. a core tablet comprising
• (i) a drug layer comprising a) an amorphous solid dispersion of apremilast and hypromellose acetate succinate (HPMCAS) and b) a water soluble polymer;
• and
• (ii) a swellable layer comprising one or more swellable polymers;
• b. a coating layer disposed on the core tablet, and
• c. a drug release orifice,
• wherein the amorphous apremilast and HPMCAS are present in a weight ratio of 40:60 to 60:40 and the oral dosage form comprises 27.5 mg apremilast or 55 mg apremilast.
