Abstract
Aspects of the present disclosure include methods for calibrating spectral data signals (e.g., flow cytometer data). Methods according to certain embodiments include irradiating a particle of a sample having a fluorophore in a flow stream with a light source, detecting light from the irradiated particle with a light detection system having a photodetector, generating spectral data signals in response to the detected light, calibrating the generated spectral data signals based on a calibration scaling factor and a calculated abundance of the fluorophore. Systems and non-transitory computer-readable storage media configured to carry out the subject methods are also provided.
Claims27
A method comprising:
- irradiating a particle of a sample comprising a fluorophore in a flow stream with a light source;
- detecting light from the irradiated particle with a light detection system comprising a photodetector;
- generating spectral data signals in response to the detected light;
- and
- calibrating the generated spectral data signals based on a calibration scaling factor and a calculated abundance of the fluorophore.

A system comprising:
- a light source configured to irradiate a particle of a sample comprising a fluorophore in a flow stream;
- a light detection system comprising a photodetector for detecting light from the irradiated particle;
- and
- a processor comprising memory operably coupled to the processor wherein the memory comprises instructions stored thereon, which when executed by the processor, cause the processor to:
- generate spectral data signals in response to the detected light;
- and
- calibrate the generated spectral data signals based on a calibration scaling factor and a calculated abundance of the fluorophore.
