Abstract
A genetic mutation to be tested is quantitatively analyzed using a nucleic acid probe. A nucleic acid fragment including a genetic mutation is detected using the nucleic acid probe in an exponential amplification phase of a nucleic acid amplification reaction.
Claims17
A method for testing a genetic mutation comprising the steps of:
- carrying out a nucleic acid amplification reaction for amplifying a region including a genetic mutation to be tested;
- detecting a target nucleic acid containing a base to be detected in the genetic mutation and a non-target nucleic acid containing a non-detection base corresponding to the base to be detected, which are contained in a reaction liquid in an exponential amplification phase of the nucleic acid amplification reaction, using a target nucleic acid detecting nucleic acid probe having a sequence complementary to a region including a base to be detected in the target nucleic acid, and a non-target nucleic acid detecting nucleic acid probe having a sequence complementary to a region including a non-detection base in the non-target nucleic acid, or a common probe having a sequence complementary to a common region that is shared by the target nucleic acid and the non-target nucleic acid and that does not overlap a region including the base to be detected or the non-detection base;
- and
- quantitatively analyzing the target nucleic acid contained in the reaction liquid.

A device for testing a genetic mutation, comprising:
- a nucleic acid amplification reaction unit adapted to carry out a nucleic acid amplification reaction for amplifying a region including a genetic mutation to be tested;
- a detection unit which is supplied with a reaction liquid from the nucleic acid amplification reaction unit, and detects at least a target nucleic acid using a target nucleic acid detecting nucleic acid probe having a sequence complementary to a region including a base to be detected in the target nucleic acid containing a base to be detected in the genetic mutation, and a non-target nucleic acid detecting nucleic acid probe having a sequence complementary to a region including a non-detection base in a non-target nucleic acid containing a non-detection base corresponding to a base to be detected, or a common probe having a sequence complementary to a common region that is shared by the target nucleic acid and the non-target nucleic acid and that does not overlap a region including the base to be detected or the non-detection base;
- and
- a liquid feeder that separates a reaction liquid in the nucleic acid amplification reaction unit, and supplies the separated reaction liquid to the detection unit.
