Abstract
Provided are gamma glutamylcysteine synthetase variants and microbial cells comprising such variants and/or expressing heterologous gamma-glutamylcysteine synthetase-glutathione synthetases, for producing increased amounts of the antioxidant peptide glutathione (GSH). Methods and compositions provided herein may be useful in baking and winemaking applications and in production of nutraceuticals, pharmaceuticals, cosmetics, plant protectants and/or animal feeds.
Claims25
A gamma glutamylcysteine synthetase comprising an amino acid sequence that is at least 90% identical to the sequence of SEQ ID NO:
- 1, wherein the amino acid sequence of the gamma glutamylcysteine synthetase comprises an amino acid substitution relative to the sequence of SEQ ID NO:
- 1 at one or more amino acid positions corresponding to positions K220, L238, C264, N277, V312, or G319 of SEQ ID NO:
- 1.
- 1 at two or more amino acid positions listed in Table 1 or Table 2.
