Abstract
The present disclosure is generally related to methods and compositions for producing heterologous globin proteins of interest in recombinant filamentous fungal cells. Certain embodiments are therefore, directed to compositions and methods for the production of globin proteins, recombinant filamentous fungal strains comprising enhanced globin protein productivity phenotypes, polynucleotides (e.g., expression constructs) encoding one or more globin proteins, industrial scale fermentation, globin protein recovery processes and the like.
Claims22
A recombinant filamentous fungal cell expressing a protease inhibitor and a heterologous globin protein, wherein the cell secretes the globin protein into the fermentation broth when fermented under suitable conditions.

A method for producing a heterologous globin protein in a filamentous fungal cell comprising:
- introducing an expression cassette encoding a globin protein and an expression cassette encoding a protease inhibitor into the filamentous fungal cell, and
- fermenting the modified cell under suitable conditions for the production of the globin protein,
- wherein the cell secretes the globin protein into the fermentation broth.
