Abstract
Aspects of the disclosure provide compounds, compositions, and methods for modulating the expression or activity of angiotensinogen (AGT). In some aspects, the compounds, compositions, and methods of the disclosure can be used to reduce the expression of AGT mRNA in a cell or animal. In some aspects, the compounds, compositions, and methods of the disclosure can be used to reduce the expression of AGT protein in a cell or animal.

A double stranded siRNA compound comprising a sense strand and an antisense strand,
- wherein the antisense strand consists of a modified oligonucleotide having a nucleobase sequence complementary to at least 21 nucleobases of positions 1815-1837 of SEQ ID NO:
- 1 and up to 23 linked nucleosides in length,
- wherein the sense strand consists of a modified oligonucleotide having a region of complementarity to the modified oligonucleotide of the antisense strand and up to 23 linked nucleosides in length,
- and wherein one or more GalNAc are attached to the 2′ or 3′ position of the ribosyl ring of the 5′ nucleoside of the sense strand.
