In vivo monitoring devices and systems for enzymes and/or analytes including devices having a reactant reservoir are provided.

A sensor for detecting enzyme activity in vivo, the sensor comprising:
- a reservoir comprising a reactant and an osmotic agent, wherein the reactant is passively released from the reservoir via diffusion;
- a membrane suitable for controlling diffusion of the reactant disposed on the reservoir;
- a working electrode disposed on the membrane, the working electrode comprising carbon nanotubes, and
- a sensing layer disposed on the working electrode, the sensing layer comprising an enzyme and capable of reacting with the reactant from the reservoir to generate a signal at the working electrode proportional to enzyme activity.

The sensor of claim 1, wherein the sensing layer further comprises a redox polymer.
