Abstract
Provided herein are orthotopic non-human mammal models of ovarian cancer and methods of generating the non-human mammal models.
Claims25
An orthotopic non-human mammal model of ovarian cancer comprising ovarian tissue with at least one cancer cell, wherein the ovarian tissue expresses an exogenous mutant PIK3CA polypeptide and an exogenous mutant KRAS polypeptide, and wherein the ovarian tissue has reduced expression of endogenous p53.
An orthotopic non-human mammal model of ovarian cancer comprising ovarian tissue with at least one cancer cell induced by expression of an exogenous mutant PIK3CA polypeptide and an exogenous mutant KRAS polypeptide, and reduced expression of endogenous p53.
A method for producing an orthotopic non-human mammal model of ovarian cancer, comprising introducing (i) a first nucleotide sequence encoding an exogenous mutant PIK3CA polypeptide, (ii) a second nucleotide sequence encoding an exogenous mutant KRAS polypeptide, and (iii) at least one inhibitory RNA targeting p53, into an ovarian tissue of a non-human mammal model under conditions effective for the formation ovarian cancer, thereby producing an orthotopic non-human mammal model of ovarian cancer.
