A method, comprising:
- administering to a subject a composition comprising a nucleic acid encoding an engineered polypeptide, wherein said engineered polypeptide comprises:
- (a) an extracellularly-oriented domain comprising an epitope capable of binding:
- (i) an antibody;
- or (ii) a peptide hormone or growth factor;
- wherein said extracellularly-oriented domain does not comprise an scFv;
- and
- (b) a transmembrane domain or membrane affinity domain capable of associating with an outer membrane of a cell, comprising a transmembrane domain or membrane affinity domain from DLL3, PSMA, SSTR2, PD-L1, EGFR, CD28, CD4, ICOS, or CD73,
- wherein (a) and (b) are heterologous to each other.
