Abstract
The invention relates to compositions and methods for the preparation, manufacture and therapeutic use of ribonucleic acid immunogenic compositions and/or vaccines comprising polynucleotide molecules preferably encoding one or more influenza antigens, such as hemagglutinin antigens, wherein the composition is frozen or lyophilized.
Claims49
A composition comprising tert-butanol and a lipid nanoparticle (LNP) comprising i) a cationic lipid, ii) a neutral lipid and/or a phospholipid, iii) a steroid, and iv) a polymer conjugated lipid.
A method for producing a composition comprising an RNA-encapsulating lipid nanoparticle (LNP) comprising the step of mixing a first composition comprising t-butanol, an ionizable lipid, a polymer-conjugated lipid, phospholipid, and a sterol with a second composition having a pH 4-6 comprising an RNA polynucleotide, aqueous solvent, and buffer, thereby producing a composition comprising the RNA-encapsulating lipid nanoparticle and t-butanol.
