Abstract
A composition for tendon and ligament regeneration includes a biologically compatible hydrogel suspension having spherical droplets. The spherical droplets have a diameter of about 100 μm to about 300 μm and a plurality of cells. The cells include marrow-derived mesenchymal stem cells (MSCs) with an average viability of about 90% or greater and cells that retain their stemness. A method for tendon and ligament regeneration includes forming the composition, purifying the composition, retrieving the composition, and administering the composition via injection into an injury site of injured tissue.
Claims20
A hydrogel-particle kit for orthopedic tissue regeneration, comprising:
- a suspension comprising hydrogel particles frozen with a cryoprotectant in a primary container, wherein the hydrogel particles comprise a hydrogel polymer lacking cell-adhesive peptide groups and viable mesenchymal stem cells encapsulated within the hydrogel particles;
- a set of instructions for a) thawing the suspension and b) injecting the hydrogel particles upon thawing into one or more of a tendon, a ligament, or a joint of a subject for promoting orthopedic tissue regeneration;
- and
- an injection device configured for injecting the hydrogel particles into an orthopedic tissue of a subject, wherein the injection device comprises a needle having a size between about 14-gauge to about 26-gauge.
A method for localized delivery and sustained release of therapeutic cells at a site of implantation, comprising:
- administering a plurality of microparticles to the site of implantation in a subject via guided implantation with noninvasive imaging, wherein each of the microparticles comprises a PEG-based hydrogel microparticle having a 3-dimensional matrix of covalently crosslinked inert polymer compounds lacking cell adhesive motifs and a desired amount of cells entrapped therein, wherein the cells have a viability of at least about 70%.
