Abstract
Described herein are methods for introducing a plurality of live cells into a fluidic device, forming a plurality of enclosures such that an enclosure of the plurality of enclosures comprises a cell of the plurality of cells, and performing one or more assays on the cell within the enclosure.
Claims30
A method, comprising:
- (a) introducing a plurality of cells to a fluidic device;
- (b) subsequent to (a), forming a plurality of enclosures, wherein a first enclosure of the plurality of enclosures comprises a first cell of the plurality of cells, wherein a second enclosure of the plurality of enclosures comprises a second cell of the plurality of cells, and wherein cytokines secreted from the plurality of cells can flow between the plurality of enclosures;
- (c) measuring an interaction between the first cell and the second cell;
- (d) subsequent to (c), releasing a nucleic acid from the first cell of the plurality of cells, wherein the nucleic acid is captured in the first enclosure;
- (e) sequencing the nucleic acid, a derivative of the nucleic acid, or a complement of a derivative of the nucleic acid;
- and
- (f) linking data generated form the sequencing in (e) to data generated from the measuring in (c).
