Abstract
Disclosed are methods for treating or preventing epilepsy in a subject in need thereof. The methods comprise administering to the subject a therapeutically effective amount of a recombinant adeno-associated viral (AAV) vector comprising a nucleic acid encoding a human claudin-5 polypeptide operably linked to an endothelial-cell-specific promoter. In certain embodiments, expression of the claudin-5 polypeptide in endothelial cells enhances or restores blood-brain barrier integrity, thereby reducing seizure susceptibility or severity.
