Abstract
The present disclosure relates to extracellular vesicles (EVs) (and their contents) from a pluripotent stem-cell derived endothelial cell product (ECP); as well as their isolation; uses and methods employing the EVs in therapy, such as in promoting angiogenesis and/or neovascularisation.
Claims17
An isolated population of pro-angiogenic and/or pro-neovasculogenic extracellular vesicles (EVs) and/or their contents thereof, obtainable from pluripotent stem-cell derived endothelial cell products.

A method of isolating EVs from endothelial cells derived from pluripotent stem cells, the method comprising:
- a) subjecting a fluid comprising EVs produced by endothelial cells derived from pluripotent stem cells to ultrafiltration, in order separate EVs from larger molecular weight material and obtain an EVs enriched fluid;
- and
- b) subjecting the EV enriched fluid to size-exclusion chromatography (SEC) in order to obtain a fluid fraction comprising isolated EVs.

A method of:
- (i) treating or preventing a cardiovascular disease;
- (ii) treating or preventing coronary heart disease;
- (iii) treating or preventing an ischemic tissue disorder;
- (iv) treating or preventing pulmonary hypertension;
- (v) modulating or promoting angiogenesis and/or neovascularisation;
- and/or
- (vi) modulating or promoting wound healing;
- said method comprising administering a subject in need thereof a miR modulator.
