Abstract
Provided herein are compositions and methods for high-throughput Primary Template-Directed Amplification (PTA) nucleic acid amplification and sequencing methods, and their applications for mutational analysis in research, diagnostics, and treatment. Further provided herein are methods for sample fixation and parallel analysis of DNA, RNA, and/or proteins from cell clusters and single cells.

A composition for cell fixation comprising:
- a cross-linking agent;
- a solvent;
- and
- an acid,
- wherein the composition is applied to a surface for fixing a cluster of one to ten cells to the surface, wherein the composition results in improved sequencing metrics.

A method of cell analysis comprising:
- a) cell fixation, wherein the cell fixation is performed using a composition comprising a cross-linking agent, a solvent, and an acid;
