Abstract
Provided herein are recombinant polynucleotides comprising a promoter, a ribosome binding site, a sequence encoding a secretion carrier, and a sequence encoding a heterologous protein. Also provided are polypeptides comprising a secretion carrier and a sequence encoding a heterologous protein. Methods of exporting heterologous polypeptides from cells are additionally provided.
Claims28
A recombinant polynucleotide comprising a promoter, a ribosome binding site, a sequence encoding a secretion carrier, and a sequence encoding a heterologous protein.
The recombinant polynucleotide of claim 6, wherein the ribosome binding site is derived from BT1311, or is RBS8 or A21 RBS.
A recombinant polypeptide comprising:
- (i) a secretion carrier comprising a positively charged region of about 3 to about 9 amino acids, a hydrophobic region of about 13 to 34 amino acids, and a lipoprotein export sequence;
- and
- (ii) a heterologous polypeptide.
