Abstract
Novel anti-effector moiety antibodies or antigen binding domains thereof and CARs that contain such effector moiety antigen binding domains, either with or without one or more booster elements, and host cells expressing the receptors, and nucleic acid molecules encoding the receptors are provided herein, as well as methods of use of same in a patient-specific immunotherapy that can be used to treat solid tumor cancers and other diseases and conditions.
Claims14
An isolated nucleic acid molecule encoding a boosted single, tandem, multi-targeting, or DuoCARs chimeric antigen receptor (CAR) comprising at least one extracellular antigen binding domain comprising a ROR1, MSLN, FOLR1, and/or CD276/B7-H3 antigen binding domain operationally linked to one or more booster elements, at least one transmembrane domain, and at least one intracellular signaling domain, which boosted single, tandem, multi-targeting, or DuoCARs CAR is encoded by a nucleotide sequence comprising SEQ ID NO:
- 151, 153, 155, 157, 159, 161, 163, 165, 167, 169, 171, 173, 175, 177, 179, 181, 183, 185, 187, 189, 191, 193, 195, 197, 226, 228, 230, 232, 234, 236, 238, 240, 242, 244, 245, 247, 249, 251, 253, or 255, or any combination thereof.

A pharmaceutical composition comprising an anti-tumor effective amount of a population of human T cells, wherein the T cells comprise a nucleic acid sequence that encodes a boosted single, tandem, multi-targeting, or DuoCARs chimeric antigen receptor (CAR), wherein the boosted single, tandem, multi-targeting, or DuoCARs CAR comprises at least one extracellular antigen binding domain comprising a ROR1, MSLN, FOLR1, and/or CD276 antigen binding domain comprising the amino acid sequence of SEQ ID NO:
- 152, 154, 156, 158, 160, 162, 164, 166, 168, 170, 172, 174, 176, 178, 180, 182, 184, 186, 188, 190, 192, 194, 196, 198, 225, 227, 229, 231, 233, 235, 237, 239, 241, 243, 246, 248, 250, 252, 254, or 256, at least one linker domain, at least one transmembrane domain, at least one intracellular signaling domain, and wherein the T cells are T cells of a human having a cancer, autoimmune, alloimmune, or autoaggressive disease or any combination thereof.
